Gerwing, Julia (University of British Columbia, Vancouver, B.C., Canada), Claude E. Dolman, and Arthur Ko. Mechanism of tryptic activation of Clostridium botulinum type E toxin. J. Bacteriol. 89:1176-1179. 1965.-The toxic peptide of trypsin activated Clostridium botulinum type E toxin was purified by chromatography through columns packed with Sephadex G-75 and G-50. The molecular weight of the active peptide was estimated to lie between 10,000 and 12,000. Amino acid analyses indicated that the active peptide had lost at least 18 of the amino acid residues present in the original protein. The active peptide and the original protein were found to have different N-terminal amino acid residues. The mechanism of tryptic activation apparently involves chiefly the removal of amino acids from the N-terminus of the toxin molecule.

Double Sandwich ELISA Modified Method for the Detection of Clostridium Botulinum Type E
Background & Objective: A very small amount of botulinum toxin can cause death and on the other hand, there is no cure for its poison other than antitoxin. Therefore, a diagnostic method that can detect very small amounts of botulinum toxin in a short time is very important. In this study, rapid and accurate detection of botulinum toxin type E has been performed with the double sandwich ELISA m...

متن کاملPurification and activation of Clostridium botulinum type E toxin.
Type E strains of Clostridium botulinum tend to produce toxin of relatively low mouse killing power (administered intraperitoneally) in relation to its high lethality per os for man. The explanation of this peculiarity seems to lie in the phenomenon of "activation," whereby type E toxin, when exposed at an appropriate pH in vitro to certain bacterial enzymes (Dolman, 1953, 1957a; Sakaguchi and ...
متن کاملAntigenicity of Clostridium botulinum type-E formol toxoid.
Tryptic activation of prototoxin of Clostridium botulinum type E increased the lethal toxicity by 250-fold, but no compatible increase in the antitoxin-combining power resulted. The formol toxoid derived from prototoxin and that from trypsin-activated 12S toxin were the same in the antigenic specificity, but prototoxin toxoid stimulated formation of a larger amount of antitoxin in guinea pigs a...
متن کاملCharacterization of Clostridium botulinum spores and its toxin in honey
Botulism is a serious paralytic disease caused by Clostridium botulinum toxin in foods. There are seven recognized serotypes of botulinum neurotoxins among which the principal prevalent types in humans include A, B and E. Infant botulism results from intestinal colonization and toxin production by C. botulinum spores in babies less than 1 year old. Honey is the most important food discriminated...
